AB OBJECTIVE: To study functions of HIV-1 Gag proteins, various in-frame
gag gene mutants were constructed and their replication properties were
examined. METHODS: Growth kinetics of mutant viruses in several cell
lines were determined by RT assays. Defective steps of the mutants in
the viral life cycle were monitored by single-round replication assays.
RESULTS: Some of the mutant viruses replicated fairly well in some
specific cell lines. In cell lines which did not support replication of
the mutants, the defects of the viruses were localized to early
infection phase in addition to late phase. CONCLUSIONS: Our results
indicate that HIV-1 Gag proteins play an important functional role,
through the interaction with a cell factor(s), at an early replication
phase. Determination of the precise defective step of the mutant viruses
is now in progress.
